No. Puzzles played in Practice Mode, Shared Puzzles, and Assignments do not affect your Global Rating or Power Score except for generic participation. They also do not contribute to the public leaderboard unless specifically played in Time Attack mode.
Assignments have private leaderboards visible only to your class and teacher. There is an overall speed leaderboard for the full assignment and individual leaderboards for each puzzle.
